摘要
以西北农林科技大学资源苗圃的18个核桃品种嫩叶为试材,利用ISSR分子标记技术对其进行了遗传多样性分析,建立了最适PCR反应体系。结果表明:从31条引物中筛选出多态性强重复性好的15条引物,共检测到132个多态位点,平均每条引物扩增出多态性位点8.8个。"西林3号"和"辽宁4号"的相似系数最大(0.7639),"西洛3号"和"新疆2号"最小(0.2500)。多态信息含量(PIC)变化范围在0.7105~0.9151,其值均大于0.5,说明15条引物扩增的位点均表现出高度多态,同时表明供试样品遗传资源非常丰富。
Taking 18 walnut varieties in nursery garden of Northwest Agricultural and Forestry University as materials,the genetic diversity of them was studied using ISSR molecular marker technology,and PCR optimum system was set up.The results showed that 15 primers were selected from 31 primers with good repeatability and strong polymorphism,132 polymorphic loci were detected;the number of the polymorphic loci of each primer averaged 8.8.The similarity coefficient between 'Xilin 3' and 'Liaoning 4' was the maximum(0.7639),'Xiluo3' and 'Xinjiang 2' was the minimum(0.2500).Polymorphic information content(PIC) ranged from 0.7105 to 0.9151,and all values were greater than 0.5,indicating that 15 primers sites showed a high degree of polymorphism,also showed that the genetic resources of these samples were very rich.
