摘要
以半夏块茎和顶芽为试材,对顶芽愈伤组织诱导、顶芽愈伤组织不定芽分化、块茎不定芽分化和不定芽诱导生根的最佳条件进行了研究。结果表明:半夏最佳顶芽愈伤组织诱导培养基为MS+6-BA 2.0mg/L+NAA 0.2mg/L;最佳顶芽愈伤组织不定芽分化培养基为MS+6-BA 1.0mg/L+NAA 0.2mg/L;最佳半夏块茎不定芽分化培养基为MS+6-BA 1.0mg/L+NAA 0.2mg/L;最佳不定芽诱导生根培养基为MS+2,4-D 2.0mg/L+KT 1.0mg/L。
Taking terminal bud and tuber as explants,the optimal conditions for inducing callus from terminal bud,differentiating of adventitious buds from callus,differentiating of adventitious buds from tuber and rooting of test-tube seedlings of Pinellia ternata(Thunb.) Breit.were studied.The results showed that the best medium to induce callus was MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,MS+6-BA 1.0 mg/L+NAA 0.2 mg/L was the best differentiation adventitious bud medium from terminal bud callus or tuber,the optimal culture medium was MS+2,4-D 2.0 mg/L+KT 1.0 mg/L for rooting.
