摘要
细胞质胸苷激酶(TK1)是嘧啶合成补救途径的关键酶,催化脱氧胸苷合成脱氧1-磷酸胸苷酸,从而参与DNA合成。TK1是一种细胞周期依赖性标志物,其在细胞周期的G1期和S期交界处开始升高,G2期最高。正常成人体内TK1含量极微,而恶性肿瘤患者由于病变细胞缺失调亡调控,DNA合成剧增,导致TK1水平的异常升高。目前TK1已被广泛用于恶性增殖疾病的诊断、疗效监测和预后判断。该文就TK1在普外科恶性肿瘤中的研究做一简要综述。
Thymidine kinase is a key cytosolic enzyme in the one-step pyrimidine salvage pathway catalysing the transfer of the terminal phosphate from ATP to 5′hydroxyl group of thymidine(dTh) to produce dThMP,which is then incorporated into DNA.TK1 is a cell cycle-dependent marker that increases markedly after the G1-S transition,and then declines rapidly in G2.The level of TK1 is minimal in the serum of healthy people,but it increases in patients with malignancies that DNA increases sharply in the diseased cells which lack apoptosis regulation.TK1 has been widely used in the diagnosis of malignant diseases,monitoring treatment and prognosis.Therefore,this review is devoted to thymidine kinase 1 in general surgery cancer.
