摘要
目的观察针刺治疗对去势大鼠偏头痛模型血清雌二醇(E2)、孕酮(P)、β-内啡肽(β-EP)、P物质(SP)水平的影响,初步探讨针刺对围绝经期偏头痛的干预作用及作用机制。方法48只雌性大鼠随机分为4组:空白对照组(即假手术组)、模型组、针刺组、雌激素组,每组12只。行相应处理和治疗30天后,采用皮下注射硝酸甘油注射液10mg/kg的方法复制偏头痛模型,造模4小时后取血,采用双抗体两步夹心酶联免疫吸附法(ELISA)测定血清E2、P、β-EP及SP含量。结果 (1)血清E2、P及β-EP含量模型组明显低于假手术组(P<0.01),而各治疗组均明显高于模型组(P<0.01),其中雌激素组高于针刺组(P<0.05)。(2)血清P含量模型组明显高于假手术组(P<0.01),而各治疗组明显低于模型组(P<0.01),其中雌激素组低于针刺组(P<0.05)。结论针刺对围绝经期偏头痛有干预作用,其可能机理是:针刺可以提高血清E2、P、β-EP含量,并降低血清SP含量,调节围绝经期内分泌紊乱,达到防治偏头痛的效果。
Objective To observe the ettect of acupuncture on the expression oi serum levets oi es- tradiol (E2), progesterone (P), beta-endorphins (beta EP), substance P (SP) in ovariectomized rat migraine model, and to preliminary discuss the action mechanism and intervention role of acupuncture to migraine menopausal transition. Methods Forty-eight female rats were randomly divided into four groups and twelve rats in each group which were control group (JiaShouShu group), model group, ac- upuncture group, estrogen group. The rats were the corresponding treatment and therapy for 30 days, adopt subcutaneous injection nitroglycerin injection 10 mg/kg method copy migraine model. After 4 hours by built mode, the serum was taken to detect the levels of estradiol (E2), progesterone (P), beta-endorphins (beta EP), substance P (SP) by enzyme-linked immunosorhent assay (ELISA) meth- od. Results The levels of estradiol (E2), progesterone (P), beta-endorphins (beta EP) in serum of model group were obviously decrease compare with which they in JiaShouShu group, and there were statistics significant differences (P〈0.01). Their levels in serum of each treatment group were obvi- ously increase compare with which they in model vels in serum of estrogen group were obviously in- crease compare with which they in acupuncture group, and there were statistics significant differences (P〈0.05). The levels of substance P (SP) in serum of model group was obviously increase compare with which it in JiaShouShu group, and there also were statistics significant differences (P〈0.01). Its levels in serum of each treatment group was obviously decrease compared with which it in model group, significant differences (P,(0.01). Its levels in serum of estrogen group was obviously decrease compared with which it in acupuncture group, and there were statistics significant differences (P〈0. 05). Conclusion Acupuncture to migraine menopausal transition have intervention role, the mecha- nism are probably, acupuncture can improve the expression of serum levels of estradiol (E2), proges- terone (P), beta-endorphins (beta EP), and can reduce the levels of substance P (SP) in serum, to adjust disorganized secretion surrounded postmenopausal period, in order to achieve the effect of migraine.
