摘要
为探讨双孢蘑菇(Agaricus bisporus)子实体发育初期的蛋白质表达变化,采用双向电泳(two-dimensional electrophoresis,2-DE)技术对双孢蘑菇As2796子实体原基与菇蕾的蛋白质组进行分析,发现14个表达差异明显的蛋白质。经质谱分析(MALDI-TOF/TOF MS)和数据库检索,鉴定到10个差异蛋白质,其中乙醇氧化酶与醇类代谢相关,磷酸烯醇式丙酮酸水合酶与能量代谢相关,蛋白酶体、5-甲基四氢三谷氨酸-同型半胱氨酸甲基转移酶、维生素B12不依赖型甲硫氨酸合成酶、1-吡咯琳-5-羧酸脱氢酶、NADP依赖的谷氨酸脱氢酶与氨基酸或蛋白质代谢直接相关,而WD40重复蛋白、肌动蛋白、热激蛋白70则与细胞内多种生物过程有关,在细胞的生长发育分化等过程中起着重要的作用;其余4个仅在双孢蘑菇蛋白质组中鉴定到功能未知的蛋白质。
Two-dimensional electrophoresis (2-DE) revealed fourteen proteins to be differentially expressed when the primordium and button stages of Agaricus bisporus As2796 fruit body formation were compared. MALDI-TOF/TOF MS analysis and database searches successfully identified ten proteins as follows: alcohol oxidase, phosphopyruvate hydratase, 5-methyltetrahydropteroyltriglutamate-homocysteine S-methyl- transferase, cobalamin-independent methionine synthase, 1-pyrroline-5-carboxylate dehydrogenase, NADP- dependent glutamate dehydrogenase, WD40 repeat protein, proteasome subunit alpha type 4, actin 1 and heat shock protein 70. The remaining four proteins have been designated as unknown proteins in the A. bisporus proteome database.
