摘要
阴离子交换层析纯化卵黄高磷蛋白,最佳条件为以NaAc/HAc缓冲液(20 mmol/L、pH 5.0并含0.3 mol/L NaCl)平衡,0.3 mol/L^0.6 mol/L NaCl缓冲液分步洗脱;产品纯度(以N/P比计)为2.78,回收率57.6%,蛋白质含量85.66%。经PVS刺激的RAW264.7细胞在考察剂量范围内不会向炎症巨噬细胞的形态转变。
Anion exchange chromatography was performed to further purify phosvitin(PVS)from egg yolk,and NaAc / HAc buffer(20 mmol/L,pH 5.0 and 0.3 mol/L NaCl) was used as starting buffer,followed by the elution with 0.3 mol/L-0.6 mol/L NaCl solution.The es-tablished method had a high recovery rate(almost 57.6 %) with N/P ratio of 2.78 and protein content 85.66 %.Simultaneously,it was found that the existence of PVS in the investigated dose range would not change cultured RAW264.7 cells to the morphology of the inflammatory macrophage.
