摘要
采用酶法水解脱脂豆饼粕制备具有生物活性的大豆小分子多肽,确定了先以粗胰酶预酶解5%豆饼粕蛋白,再以中性微生物蛋白酶进一步水解提取制备大豆小分子肽(分子量300 u^700 u)后,通过动物实验研究该大豆小分子肽的抗疲劳和抗氧化等生物学功能效果。采用昆明种小鼠,实验组小鼠分低,中,高(3、6、9 g/kg/d)3个剂量组灌胃大豆小分子肽25 d,测定小鼠的力竭游泳时间,采用Wister大白鼠,实验组大鼠同样分3个剂量组灌胃大豆小分子肽50 d(剂量相同),测血红细胞和肝脏中超氧化物歧化酶(SOD)活力,采用老年雄性Wister大鼠20只,随机分为老年对照组10只,老年实验组10只,老年实验组灌服大豆小分子肽,按6.0 g/kg/d剂量,老年对照组灌服同体积的生理盐水。第60天末取样测定血红细胞和肝脏中丙二醛含量、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性。大豆小分子肽(分子量300 u^700 u)的中;高剂量组能明显延长小鼠负重游泳时间。并且高剂量组的大鼠的血红细胞的超氧化物歧化酶(SOD)活力明显高于对照(P<0.05),而肝脏组织SOD酶活性有升高的趋势,但无统计学意义(P>0.05)。血红细胞与肝脏组织丙二醛含量有减低趋势,但无统计学意义(P>0.05);GSH-Px酶活性升高(P<0.05)。大豆小分子肽(分子量300 u^700 u)具有一定的一定增强小鼠抗疲劳能力和提高老龄大鼠体内抗氧化酶活性、抑制脂质过氧化的作用。
Abstract: The new extraction technique of soybean bioactive minor peptides from defatted soy flakes ydrolysized by enzymatic method. Soybean minor peptides (MW300 u-700 u) were obtained. To study and discuss the anti- fatigue effects and biological mechanism of Soybean minor peptides and the anti-oxidation effects and the antiaging effect. Soybean minor peptides was administered to mice in treated group for 25 days by 3.0,6.0,9.0 g/ kg/d respectively, and the changes of exhaustive swimming time and biochemical index were measured : blood and liver activities of superoxide dismutase (SOD) of Hb and liver in the mice and. Twenty Wister old rats were randomly divided into 2 groups:old control group and testing old group with 10 rats in each group.Testing old group were perfused with Soybean minor peptides (6.0 g/kg/d)for 60 days.The rats old control group were fed with water in the same volume.At the end of the 60th days and measured the activities glutathione peroxidase (GSH-Px) and the concentration of malondialdehyde(MDA)in Hb and liver. Compared with control group, the exhaustive swimming time of the administered mice in treated group were significantly longer (P〈0.05) ,while the SOD activities of Hb in mice obviously increased and compared with control group in the old control group, the concentrations of MDA in Hb and liver had also a increase tendency,the the activities of GSH-px had a increase tendency, (P〈0.05). The Soybean minor peptides (MW300 u-700 u) have a potential anti-fatigue effect and increasing the activity of antioxidant enzymes and decreasing lipid peroxidation.
