利用高效液相色谱法建立人体内药代动力学

摘要 采用高效液相色谱法测定血清中利复星药物浓度 ,8例肺结核患者口服 40 0mg利复星后 ,血峰浓度于1 2 2h到达 ,为 4 52± 0 72mg/L ,一次给药 2 4h后最低血药浓度为 0 52± 0 2 6mg/L ,AUC为 48 35± 5 38mgh/L ,半衰期为 6 4 9± 1 70h ,表观分布容积V/FC为 77 4 8± 8 33/L ,证明患者 2 4小时体内血药浓度维持在最低杀菌浓度MBC之上。 In patients with pulmonary tuberculosis by given 400mg orally in 8 patients. Concentrations of Levofloxacin methane sulfonic acid in serum were measured by HPLC. The results showed that a one compartment model with 1 st order absorption are fitted for LFX MAS, the average values of peak time is 1.22h, maximal concentrations of LFX MSA in serum is 4.52±0.72mg/L, the minial concentrations of LFX MSA after a singal does is 0.52±0.26mg/L, the average area under the curve is 48.35±5.38hmg/L, the half life time of elimination is 6.49+1.70h, the apparent volume of distribution is 77.48±8.331, the concentrations of LFX MSA is serum around 24 hours would be maintained above the minimal concentrations of bactericide.
