他克莫司滴眼液对损伤角膜上皮AQP3、5表达的影响 被引量:1

Influence of Tacrolimus Eye Drops on expression of aquaporin protein 3 and 5 in injuried corneal epithelium

摘要 目的通过研究他克莫司(tacrolimus)滴眼液(0.1%FK506)在损伤角膜上皮愈合过程中水通道蛋白(AQP)3、5的表达及其作用机制,为研制角膜上皮创伤修复的有效治疗药物提供理论依据。方法采用机械法刮除小鼠角膜中央上皮的方法建立模型,观察角膜上皮修复情况。取损伤若干小时用药后大鼠角膜进行AQP3、5免疫组化染色,观察AQP3、5表达情况。结果角膜上皮基底细胞层可见AQP3表达,并且实验组表达比对照组更高,表达高峰出现在损伤后18 h。角膜上皮损伤后用0.1%FK506滴眼,伤后6 h实验组愈合面积与对照组比较,差异无统计学意义,而损伤后12、18、24 h比较,差异有统计学意义(P<0.05)。上皮全层可见AQP5表达,随着角膜的愈合,其表达量降低。实验组6、12、18 h表达量与对照组比较,差异有统计学意义(P<0.05)。结论应用0.1%FK506滴眼,可通过增加AQP3表达而加快小鼠角膜上皮损伤的修复;而AQP5表达量则随角膜愈合时间延长而降低。 Objective To investigate the influence of Tacrolimus Eye Drops(0.1%FK506) on the expression of aquaporin protein 3 (AQP3) and aquaporin protein 5 ( AQP5 ) during the healing process of corneal epithelial injury to provide the theoreti- cal basis for researching and developing the effective curative drugs in the repairment of corneal epithelial injury. Methods The mechanical curettage of central corneal epithelium in mice was adopted to establish the model for observing the repair of coneal epithelium. The mice corneas were taken after some hours of O. 1%FK506 treatment for conducting the immuohistochemical stain- ing and observing the expression of AQP3 and AQP5. Results AQP3 expression could be seen in the basal cell layer of cornea epithelium,which was higher in the experimental group than in the control group,and the expression peak occurred after 18 b of injury. After the use of 0.1%FK506 in corneal epithelial injury, the comparison of epithelial healings areas showed no statistical difference after 6 h of injury between the experimental group and the control group, while the differences after 12,18,24 h of in- jury showed statistical signiflcance(P〈O.05 ). The AQP5 expression could be seen on the whole epithelial layers ,and the expression quantity of AQP5 was decreased with corneal healing. The differences among comparisons after 6,12,18 h of injury showed statisti cal significance (P〈0.05). Conclusion 0.1%FK506 can accelerate the epithelial wound repair in mice by increasing the AQP3 expression and the AQP5 expression is decreased with the extension of corneal healing time.
