呋喃西林代谢物在凡纳滨对虾体内代谢及残留 被引量:6

Pharmacokinetics and residues of metabolite of nitrofurazone(semicarbazide)in Penaeus vannamei

摘要 为研究呋喃西林代谢物氨基脲(semicarbazide,SEM)在凡纳滨对虾体内代谢及残留消除。在试验水温(28±2)℃下,以30 mg/kg的剂量对凡纳滨对虾多次药饵给药后,采用高效液相色谱-串联质谱法测定血淋巴和组织中药物浓度,并通过WinNonlin 5.2药代动力学软件及Excel2010软件进行数据分析。经房室模型分析,SEM在血淋巴中吸收、分布、消除半衰期(t1/2K01、t1/2α、t1/2β)分别为0.695,2.567,47.603h。经非房室模型的统计矩原理分析,SEM在血淋巴、肌肉、鳃、肝胰腺和肠的平均驻留时间MRT分别为34.424,49.281,39.178,32.390,35.170h。停药2d后药-时曲线经Excel拟合,SEM在血淋巴、肌肉、鳃、肝胰腺和肠的消除半衰期t1/2β大小为肌肉11.75d>血淋巴8.88d>鳃8.66d>肝胰腺5.37d>肠线4.71d。结果表明,SEM在凡纳滨对虾体内吸收快、分布广,但消除缓慢。 Abstraet:Pharmaeokinetics and residues of metabolite of nitrofura- zone(semicarbazide, SEM) were investigated in Penaeus vannamei,under experimental field conditions at (28±2)℃ after oral multi- dose administration (30 mg/kg). The concentrations of SEM in plas- ma and tissues were determined by using high performance liquid chromatography tandem mass spectrometry(LC--MS/MS) after liq- uid-liquid extraction and analyzed by software of WinNonlin5.2 and Excel 2010. Compartmental analysis showed that the half-life of ab- sorption (t1/2KO1), distribution (t1/2α) and elimination (t1/2β) were 0. 695, 2. 567 and 47. 603 h, respectively. Non-compartmental anal- ysis based on statistical moment theory showed that the mean resi- dence time (MRT) of SEM in plasma, muscle, gill, hepatopancreas and intestine were 34. 424, 49. 281, 39. 178, 32. 390 and 85. 17 h, respectively. At the 2th day post-treatment, the SEM half-lives of elimination phase in muscle, plasma, gill, hepatopancreas and intes- tine were 11.75, 8.88, 8.66, 5.37 and 4.71 d, respectively. The results of experimentation indicated that absorption and distribution of SEM in Penaeus vannamei were fast and wide, but eifmination was SLOW.
