摘要
研究色胺盐酸盐的合成方法并对其工艺进行优化。以吲哚为原料,经Vilsmeier反应、缩合、还原3步反应得到目标化合物,通过1HNMR、13CNMR确认产物结构,并通过HPLC与标准品对照测得其纯度。新的工艺对每步反应条件进行了优化,色胺盐酸盐的总产率从文献报道的20%~63%提高至80%,纯度达99%。该方法具有原料廉价易得、条件温和、操作方便、收率好的优点。
Synthesis of tryptamine hydrochloride and the optimization of its process were studied.The target compound was prepared using indole as start material via three steps: Vilsmeier reaction,condensation and reduction.The structure of the product was confirmed via 1 HNMR and 13 CNMR,and the purity was measured via HPLC by contrast with standard substance.By optimizing each step,the total yield of tryptamine hydrochloride was raised to 80% from that of previously reported 20% ~ 63%,and its purity reached 99%.The process has several advantages such as low cost,easy availability of starting material,mild conditions,simplified operation and high production yield.
