摘要
目的研究hIL-24(人白细胞介素24)对MDA-MB-231乳腺癌细胞的生长抑制作用。方法将pDC316-hIL-24-EGFP转染人乳腺癌MDA-MB-231细胞,用RT-PCR法、Western blot法检测IL-24基因在肿瘤细胞中的表达。结晶紫染色法检测IL-24基因的表达对乳腺癌细胞的生长抑制。RT-PCR检测侵袭、转移相关基因的转录。ELISA法检测VEGF、Ang-1的分泌水平。结果 IL-24基因可在MDA-MB-231细胞中成功转录及表达,并对乳腺癌细胞增殖有明显抑制作用。能明显下调MMP-2、MMP-9的转录。VEGF、Ang-1表达下降。结论 IL-24对乳腺癌细胞有明显的抑制生长、侵袭转移、血管生成的作用。
Objective To explore the effect of IL-24 gene on proliferation of human breast cancer cell line MDA-MB- 231. Methods The recombinant shuttle vector pDC316-hlL-24-EGFP was transiently transfected into human breast cancer cell line MDA-MB-231 with lipofectamine. Both IL-24 mRNA and protein were detected by RT-PCR and West- ern blot respectively. The inhibition of cell proliferation was evaluated by crystal violet staining assay. The expression level of VEGF and Ang-1 gene were detected by ELISA. Results IL-24 was proved successfully transcribed and ex- pressed in MDA-MB-231 cells. The IL-24 expression inhibited MDA-MB-231 cells proliferation. The mRNA expression of MMP-2 and MMP-9 reduced in IL-24 group compared with PBS+ pDC316-EGFP group. The expression of VEGF and Ang-1 were down. Conclusion IL-24 gene overexpressed was able to effectively inhibit the proliferation,invasion, metastasis and angiogenesis of breast cancer cells.
