摘要
目的研究长沙地区汉族人群人类血小板特异性抗原HPA1-17基因多态性。方法采用PCR-SSP方法对长沙市血液中心618名汉族无偿献血者进行HPA1-17系统基因分型。结果长沙地区618名汉族无偿献血者HPA1a基因频率为99.3%,HPA2a基因频率为96.8%,HPA3a基因频率为58.7%,HPA4a基因频率为99.4%,HPA5a基因频率为98.9%,HPA6a基因频率为98.9%,HPA15a基因频率为53.6%。结论长沙地区汉族人群HPA1-17的基因检测填补了地方空白,为建立血小板库奠定了基础,也为临床提供同型血小板输注提供可能。
Objective To investigate the genetic polymorphism of HPAI-17 antigens in Changsha Han population. Methods A total of 618 samples from unrelated platelet donors of Changsha were genotyped by Polymerase Chain Re- action with Sequence-Specific Primers （PCR-SSP）. Results The allele frequencies of HPA-la, 2a, 3a, 4a, 5a, 6a, 15a were 0. 993,0. 968,0. 587,0. 994,0. 989,0. 989,0. 536, HPA-Ta-17a（exclude 15a） were 1. 000, HPA-bw alleles were not detected. Conclusion The genotype of HPA can improve the safety and curative effect of the platelet transfusion. Genotyping can be used in HPA crossmatch and the diagnosis of alloimmune platelet disease.
