摘要
从吉林某猪场采集疑似患断奶仔猪多系统衰竭综合征死亡的猪体病料,进行猪圆环病毒2型(PCV-2)的分离;分离培养物经荧光标记的PCV-2特异抗体染色鉴定,并用PCV-2全基因组的特异性引物扩增,进行序列测定和分析。结果表明,从采集病料中成功分离出一株猪圆环病毒2型,并将此吉林分离株命名为PCV-2-JL11S,对其进行全基因测序,拼接结果显示,基因组全长为1 767bp;将分离株病毒的全基因组序列与14株GenBank已登录病毒进行比较,结果显示,核苷酸同源性最高可达99.2%;全基因序列系统进化树结果表明,分离株与HQ395057关系最近,而与HQ395037关系较远。上述结果表明,PCV-2吉林分离株存在一定的基因变异,而变异的发生原因尚需深入研究。
Clinically suspected post-weaning multisystemic wasting syndrome (PMWS)samples were collected from a pig farm in Jilin,and the isolation of porcine circovirus type 2 was performed. The isolated culture was characterized by fluorescent labeled PCV-2-specific antibody staining, PCR amplification and sequence analysis were conducted with PCV-2 genome specific primers. The results showed that a strain of porcine circovirus type 2 was isolated successfully, named PCV-2-JLllS, and its whole genome is 1 767 bp, with the highest nucleotide homology with the sequenced strains up to 99.20/00. Phylogenetic analysis showed that the relationship of PCV-2 isolates is closest with HQ395057, while distant with HQ395037 ; These results indicated that there is a certain genetic variation in PCV-2 Jilin isolate, and the cause of variation will take in-depth study.
