摘要
目的探讨抑癌基因maspin在胃癌细胞株SGC-7901生长和凋亡过程中的生物学作用。方法构建重组质粒pCD-NA3.1-maspin,转染胃癌细胞株SGC-7901,采用逆转录-聚合酶链反应(RT-PCR)和免疫细胞化学检测maspin基因表达,MTT法检测细胞增殖,原位末端标记法(TUNEL)检测细胞凋亡。结果重组质粒pCDNA3.1-maspin导入SGC-7901细胞株后,maspin的mRNA和蛋白表达明显增强(P<0.05),胃癌细胞株SGC-7901增殖受到抑制,并出现明显的细胞凋亡。结论 maspin基因可以抑制胃癌SGC-7901细胞增殖,并诱导细胞凋亡。
Objective To investigate the biological effect of maspin gene on growth and apoptosis of gastric carcinoma cell SGC- 7901 in vitro. Methods pCDNA3.1-maspin vector was constructed and transfected into SGC-7901 cells. Maspin gene expression was detected with RT-PCR and cell immunochemistry. Cells growth rate was detected with MTT,and cell apoptosis was tested by TUNEL. Results After transfected with pCDNA3. 1-maspin, both RNA and protein levels of maspin increased obviously(P( 0. 05). The proliferation of gastric cancer cells SGC-7901 was suppressed obviously, and cell apoptosis is obvious. Conclusion Maspin gene can suppress the proliferation of gastric carcinoma ceils arid induce cell apoptosis.
