丹参注射液对肺栓塞大鼠内皮素及P选择素表达水平的影响

Effects of Danshen Injection on the Expression of P-Selectin and Endothelin of Rat with Pulmonary Thromboembolism

目的观察丹参注射液对大鼠急性肺栓塞后肺损伤的作用,对肺栓塞大鼠血浆内皮素(Endothelin,ET-1)含量、肺血管内皮细胞P选择素表达水平的影响。方法建立大鼠急性肺栓塞模型,随机分为对照组、栓塞组和丹参组,丹参组于制备肺栓塞模型成功后立即腹腔注射丹参注射液,模型组给予等容量的生理盐水,取肺组织和血清,采用放射免疫分析法测定肺栓塞不同时间点血液中ET-1的变化情况,Western blot法检测肺血管内皮细胞P选择素蛋白的表达。结果血浆中ET-1含量及肺血管内皮细胞P选择素蛋白表达水平在栓塞后开始即增高(P<0.05),丹参组经治疗后血浆ET-1含量、肺血管内皮P选择素蛋白表达水平均下降与栓塞组比较有显著性差异(P<0.05)。结论丹参注射液可能通过降低肺栓塞时ET含量及P选择素表达水平,减轻炎症反应,对急性肺栓塞后肺组织损伤有一定的保护作用。

Objective To observe the effects of Danshen injection on the expression of P- selectin and ET- 1 of PTE model rats. Methods Pulmonary thromboembolism （PTE） model was established first. Then Rats were randomly divided into control group, PTE model group and Danshen-treated group. Danshen-treated group was given Danshen intraperitoneal injection immediately after the PTE model group established success- fully. PTE model group was given saline,the lung tissue and serum were used to check blood ET-1 changes by the radioimmunoassay determination of pulmonary embolism at different time points, and P-selectin protein ex- pression by Western blot assay. Results Plasma ET-1 in pulmonary vascular endothelial cells and the expres- sion levels of P seleetin protein both increased after embolization（ P 〈 0.05 ）, plasma ET-1 levels in pulmonary vascular endothelial and the expression levels of P- selectin protein of Danshen group both declined after treat- ment. The differences between PTE model group and Danshen-treated group were statistically significant（ P 〈 0.05 ）. Conclusion Danshen injection may reduce pulmonary embolism ET content and P-selectin expres- sion levels, so it has a certain protective effect to the lung tissue.
