摘要
采用高糖预处理和高糖培养完全可以遏制体细胞的分裂和促进高比例花粉愈伤组织的形成。分化培养中及时由高激素水平转移到低激素水平下培养,可以获得1.96%根芽同时分化的完整植株。由这类植株的根尖染色体鉴定得到 n=10的单倍体,并移栽成活。在适宜的激素配比下,添加生长延缓剂嘧啶醇,可使带根茎的肉质根分化率提到87%以上,移栽成活率近100%。同时对根的细胞分化和形态发生作了细胞和组织学研究。
Asparagus pollen-originated calli were initiated on the Murashige and Skoog basal medium supplemented with 2mg/L NAA and 1 mg/LBA with high sucrose concetration and low temperature pretreatment.1.96 percent of the calli developed shoots and roots as well.To check the chromsome number of the root tips of these plantlets,we obtained the haploids (n=10) and transplanted successfully.Shoot tips were cultured on the half strength MS (Except Fe Na_2 -EDTA) medium with 0.5 mg/L NAA,0.125 mg/L 2ip,and 0.5 mg/L ancymidol, the rooting percentage can be raised to 87%,and nearly 100% of these plants survived transplanting,Moreover,we underwent the cytological and histologic- al observation of cell differentiation and organogenesis.
