摘要
目的用ROCK特异性抑制剂Y-27632处理Hep-2细胞系后,探讨ROCK信号转导通路对细胞的生长、侵袭和迁移的影响。方法商品化细胞系Hep-2经Rho激酶ROCK抑制剂Y-27632处理后,分别在光镜下观察细胞形态学变化,通过迁移实验和Transwell matrigel侵袭实验及MTT实验,观察细胞运动、迁移增殖能力的变化。结果经Y-27632处理后,Hep-2细胞的形态明显变化,细胞的侵袭能力明显下降,运动能力下降。结论 ROCK信号转导通路参与Hep-2细胞体外运动、侵袭、迁移的调节。ROCK特异性抑制剂Y-27632可降低Hep-2细胞的侵袭能力,干扰ROCK有望作为喉癌靶向治疗的新靶点,为喉癌的靶向治疗或新基因治疗带来希望。
Objective To explore the influence of ROCK signaling pathway on the growth, invasion and migration of Hep - 2 cells by using ROCK kinase inhibitor Y - 27632 to treat Hep - 2 cell lines. Methods We used the ROCK kinase inhibitor Y - 27632 to deal with the Hep - 2 cells and observe the Hop - 2 cells morphological changes. We used the Transwell and MTT to observe the cell motility,migration, and proliferative capacity. Results The Hep -2 cells had obvious morphological changes after Y -27632 treatment. Their in- vasive ability and motility decreased significantly. Conclusion ROCK signaling pathway involved in the regulation of Hep2 cell move- ment,invasionand migration. ROCK - specific inhibitor Y - 27632 can reduce the invasive ability of Hep - 2 cells. It may he a new target of laryngeal cancer targeted therapy to interfere with ROCK signaling, and bring hope for targeted therapy of laryngeal cancer.
