Proportional Characteristics of Hyperactivation in Human Sperm

摘要 目的研究正常人精子超激活运动的比例特征。方法 11例常规检查正常的精液,通过Percoll分离处理再分别在获能条件下培养3和5h,利用计算机辅助精子分析仪对人精子直线速度(VSL),曲线速度(VCL),平均路径速度(VAP),线性[LIN,(VSL/VCL)×100],头部侧振幅(ALH)和精子超激活(%)进行检测分析。结果在获能条件下培养3h时,精子超激活运动比例的均值为25.3%,标准差13.2%,中位数为27.0%,95%可信区间为16.4%~34.1%;在5h时,比例的均值为18.9%,标准差14.3%,中位数为18.0%,95%可信区间为9.3%~28.5%;两者服从正态分布。在获能培养3~5h过程中,人精子超激活运动的比例随时间延长呈降低趋势,但统计学差异不显著。结论人精子在获能培养3h和5h时超激活运动比例的均值分别25.3%和18.9%,95%可信区间分别为16.4%~34.1%和9.3%~28.5%,并且比例随时间的延长而呈降低趋势。 Objective To investigate the proportional characteristics of hyperactivation in human sperm. Methods The normal se- men was separated by the discontinuous Percoll gradients hefore human sperm were cultured under capacitation conditions for 3h or 5h. Straight line velocity ( VSL), curvilinear velocity ( VCL), average path velocity ( YAP), straightness ( STR, VSL/VAP by multiple 100) , linearity (LIN, VSL/VCL multiplied by 100) , amplitude of lateral head displacement (ALH) , and the percent of hyperactivation was assessed by computer assisted sperm analyzer ( CASA). Results When human sperm were cultured under capacitation conditions for 3 hours, the mean percent of hyperactivation was 25. 3% , standard deviation 13. 2% , median 27. 0% , 95% confidence interval 16.4% - 34.1% ; for 5 hours, the mean percent of hyperactivation was 18.9% , standard deviation 14.3% , median 18.0% , 95% confi- dence interval 9.3% -28.5%. The percent of hyperactivation in both groups obeyed the normal distribution. Furthermore, the percent of hyperactivation was decreasing from 3h to 5h, but the difference between two groups was not statistically significant. Conclusion The mean percents of hyperactivation in human sperm cultured under capacitation conditions for 3h and 5h are 25.3% and 18.9% , respective- ly; the 95% confidence intervals are 16.4% -34.1% and 9.3% -28.5% , respectively, and the percent of hyperactivation is decrea- sing with increased time.
