摘要
目的研究酪氨酸激酶受体RON在子宫内膜腺癌中的表达及其与临床病理参数的关系。方法采用原位杂交和免疫组化方法分别检测20例正常子宫内膜组织、20例不典型增生子宫内膜组织、60例子宫内膜腺癌组织之间的RON mRNA和蛋白表达。结果不同子宫内膜组织间原位杂交单位面积平均光密度(OD)值分别为:正常子宫内膜0.422±0.132、不典型子增生宫内膜0.700±0.143、子宫内膜腺癌1.038±0.164。RON在正常子宫内膜和不典型增生子宫内膜与子宫内膜腺癌比较有统计学意义(P<0.01),但不典型增生子宫内膜与正常子宫内膜无统计学意义(P=0.631)。不同子宫内膜组织间免疫组化OD值分别为:正常子宫内膜0.474±0.223、不典型增生子宫内膜1.078±0.086、子宫内膜腺癌1.217±0.107。RON在正常子宫内膜、不典型增生子宫内膜与子宫内膜腺癌比较有统计学意义(P<0.01),不典型增生子宫内膜与正常子宫内膜相比较也有统计学意义(P=0.015)。RON mRNA和蛋白表达与子宫内膜腺癌临床分期、组织学分级及淋巴结转移有关(P<0.05),RON mRNA还与肌层浸润深度有关(P<0.01)。结论 RON的过度表达参与子宫内膜腺癌的发生、发展过程。
Objective To investigate the relation ship between tyrosine kinase receptor RON expression and clinicopathological pa- rameters, and observe the expression of RON in endometrial adenocarcinoma( EC). Methods The expression of RON mRNA and protein was studied in 20 cases of normal endometrium ,20 cases of atypical endometrial and 60 cases of endometrial adenocarcinoma by in situ hy- bridization and immunohistochemistry method. Results Average optical density (OD) of per unit area of in situ hybridization was 0. 422 ± 0. 132 in normal endometrium, 0. 700 ± 0. 143 in atypical endometrial and 1. 038 + 0. 164 in endometrial adenocarcinoma. It prompted that the expression of RON mRNA in normal endometrium and atypical endometrial each had statistical significance to that in endometrial adenocarcinoma (P 〈 0.01 ). But the expression of RON mRNA in normal endometrium had no statistical significance to that in atypical endometrial(P = 0. 631 ). Average optical density of per unit area of immunohistochemistry was 0. 474 ± 0. 223 in normal endometrium, 1. 078 ± O. 086 in atypical endometrial and 1. 217 ± 0. 107 in endometrial adenocarcinoma. It revealed that the expression of RON protein in normal endometrium and atypical endometrial each had statistical significance to that in endometrial adeuocarcinoma ( P 〈 0.01 ) , and there was statistical significance between normal endometrium and atypical endometrial as well(P = 0. 015). The statistical evaluation ex- posited that the expression rates of RON mRNA and RON protein had close relations with clinical stage, histologic grade as well as lym- phatic metastasis ( P 〈 0.05 ), had nothing to do with ages, menostasis and the depth of Infiltration in Musclar layer ( P 〉 O. 05 ), al- though the expression of RON mRNA was relevant to the depth of infiltration in Musclar layer ( P 〈 0.01 ). Conclusion Over - expression of RON plays a role in the occurrence and development processes of endometrial adenocarcinoma.
