百会穴局部腺苷A1受体对电针诱导脑缺血耐受的作用 被引量:6

Local adenosine A1 receptors of baihui acupoint mediate cerebral ischemia tolerance induced by Electroacupuncture

摘要 目的观察百会穴局部腺苷A1受体(A1R)是否介导电针诱导的脑缺血耐受。方法将60只sD大鼠按随机区组的方法分成5组:假手术组(S组);模型组(M组);电针组(E组);穴位注射A1R激动剂组(CCPA组);穴位注射二甲基亚砜组(DMSO组),每组12只。采用大脑中动脉阻塞(MCAO)法建立大鼠局灶性脑缺血再灌注损伤模型。E组在造模前2h电针预处理大鼠百会穴;CCPA组和DMSO组分别在造模前2h百会穴注射CCPA(0.1mmol/L)、DMSO各20山。再灌注24h时观察大鼠的神经行为学后立刻处死大鼠,各组6只大鼠行Trc染色评估脑梗死体积,另外各组剩余6只大鼠行免疫印迹(Western blotting)法检测脑组织中Bc1-2蛋白表达量。结果E组与M组相比较神经行为学表现有改善;脑梗死体积从61.72%±7.12%(M组)减小为22.37%±4.42%(E组);Bcl-2蛋白表达量从0.52%±0.19%(M组)增加为1.07%±0.41%(E组),差异都具有统计学意义(P〈0.05)。CCPA组与M组和DMSO组相比较行为学有改善;脑梗死体积分别为61.72%±7.12%(M组),56.90%±4.7%(DMSO组)减小为25.12%±4.17%(CCPA组);Bc1-2蛋白表达量分别为0.52%±0.19%(M组),0.56%±0.17%(DMSO组)增加至0.92%±O.37%(CCPA组),差异均有统计学意义(均P〈0.05)。E组和CCPA组行为学表现、脑梗死体积分别为22.37%±4.42%(E组)和25.12%±4.17%(CCPA组),和Bcl-2蛋白表达量分别为1.07%±0.41%(E组)和0.92%±0.37%(CCPA组),差异均无统计学意义。结论电针可以诱导MCAO大鼠的脑缺血耐受;百会穴局部A1R可能介导了电针预处理MCAO大鼠的脑缺血耐受作用。 ischemia tolerance Objective To investigate whether induced by el local A1R of Baihui acupoint mediate cerebral (EA). Methods Sixty SD rats were randomly divided into five groups, i.e., the sham-operation (S) group, the model group (M), the electroacupuncture (E) group, the CCPA group and the DMSO group. The focal cerebral ischemia/ reperfusion model was established by middle cerebral artery occlusion (MCAO) in rats. Rats in the E group were received EA pretreatment baihui acupoint at 2 h before established MCAO. The rats in DMSO group and the CCPA group were injected with DMSO (20 td) and CCPA (0. 1 mmol/L) 20into Baihui, respectively,at 2 h before established MCAO. After 24 h reperfusion, the rats' behavior, cerebral infarct volume, the cerebral Bcl-2 protein expression were assessed. Results Compared with M group, the rats' behavior was improved, the cerebral infarct volume was decreased and the Bcl-2 protein expression was up- regulated (P 〈0. 05) in the E group. Compared with M and DMSO group, the rats' behavior was improved, the cerebral infarct volume was decreased and the Bcl-2 protein expression was up-regulated ( P 〈 0. 05 ) in the CCPA group. There were no statistical differences between CCPA and E group. Conclusions EAinduced cerebral ischemia tolerance. Local AIR of Baihui acupoint possible mediate cerebral ischemia tolerance induced by Electroacupuncture.
