胃癌癌胚抗原相关黏附分子6表达上调并促进其迁移和侵袭

CEACAM 6 expression upregulated and it enhances tumor cell migration and invasion in gastric cancer

目的:探讨癌胚抗原相关黏附分子6(carcinoembryonic antigen-related cell adhesion molecule 6,CEACAM6)在胃癌中表达的临床意义及其生物学功能。方法:采用实时定量PCR检测51例胃癌组织及其对应癌旁非肿瘤组织CEACAM6的mRNA表达,分析其与临床参数之间的关系;CEACAM6 siRNA转染细胞株SNU-16、MKN-28,下调其CEACAM6表达,采用transwell小室和Matrigel胶等方法检测下调CEACAM6表达后胃癌细胞迁移和侵袭能力的变化。结果:58.8%(30/51)胃癌病例的肿瘤组织CEACAM6表达高于对应的癌旁非肿瘤组织,癌旁非肿瘤组织CEACAM6基因相对表达量中位数7.04低于肿瘤组织表达量中位数19.10(P=0.021),肿瘤浸润越深,其CEACAM6表达水平越高,差异具有统计学意义(P=0.017);MKN-28、SNU-16两株细胞株通过siRNA干扰技术下调CEACAM6表达后,迁移和侵袭能力分别下降57.11%、85.54%和52.08%、90.77%。结论:在半数以上胃癌病人中,肿瘤组织中CEACAM6表达高于其对应癌旁组织,肿瘤组织中CEACAM6表达与浸润深度呈正相关,下调CEACAM6表达后能明显抑制胃癌细胞的侵袭和迁移,提示其在胃癌发生发展过程中起着促癌基因的作用。

Objective To investigate the significance and biological function of carcinoembryonic antigen-related cell adhesion molecule 6 （CEACAM6） in gastric carcinomas. Methods CEACAM6 expression level were measured by quantitative realtime PCR in 51 gastric cancer samples, and the relationships between CEACAM6 expression level and clinical pathological features of the samples were analyzed. Downregulation of CEACAM6 expression in SNU-16 and MKN-28 cell lines was achieved by siRNA interference. The migration was investigated by transwell chambers, and invasion by matrigel, respectively. Results CEACAM6 expression was higher in tumor tissues than that in the matched adjacent normal tissues in 58.8% samples. The median of CEACAM6 mRNA expression level was higher in tumor tissues than in matched non-tumor tissues （19.10 vs 7.04, P=0.021）. CEACAM6 expression level was positively correlated with the depth of tumor invasion. After downregulation of CEACAM6 by siRNA interference, migration and invasion decreased by 57.11% and 85.54% in MKN-28 cells and 52.08% and 90.77% in SNU-16 cells, respectively. Conclusions CEACAM6 expressed higher in tumor tissues and its expression level was positively correlated with the depth of tumor invasion. The invasion and migration ability of gastric cancer cells was inhibited when CEACAM6 expression was downregulated. CEACAM6 may function as a pro-oncogene in the development of ~astric cancer.
