摘要
目的:建立栀子苷的定向分离模式,提取栀子药材中的栀子苷。方法:依托以Lipid A为靶点的抗LPS药物筛选平台,应用大孔吸附树脂、膜分离和HPLC等技术对栀子水提液进行针对Lipid A的逐级分离纯化。结果:从栀子水提液中分离出活性化合物栀子苷,其与Lipid A的特异性结合值为112 arc seconds。结论:本研究所建立的栀子苷的定向分离模式,目的性强、效率高,可用于天然药物中抗LPS活性物质的富集与分离纯化。
Objective: To establish the method of directional isolation and extraction of jasminoidin from aqueous extract of Gardenia jasminoides Ellis (GjE). Methods : Based on the biosensor technology platform for screening anti - LPS compound which targeted as Lipid A, the aqueous extract of GjE was isolated and purified gradually by using macroporous adsorptive resins, membrane separation and HPLC technology in turn. Results:The jasminoidin was attained form aqueous extract of GjE, the value of specific binding to Lipid A was 112 arc/s. Conclusion: The established directional isolation mode of jasminoidin is more effective than that of traditional method, which is applicable for enriching and purifying anti -LPS active compound from natural medicine.
