E-cadherin基因启动子C-160A SNP与胃癌的相关性 被引量:4

摘要 目的探讨E-cadherin基因启动子区C-160A与胃癌易感性之间的联系。方法通过聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法,分析150例健康人群(对照组)和150例胃癌患者(胃癌组)的E-cadherin启动子区-160位点SNP情况。结果胃癌组E-cadherin基因启动子区-160位点基因型和等位基因型分布与对照组比较差异具有统计学意义(P<0.05)。根据吸烟状况、上消化道恶性肿瘤家族史和术后病理分期分层分析发现,无论吸烟与否,在胃癌患者和健康个体中,E-cadherin-160基因型及等位基因型分布均有显著性差异(P<0.05),而无论吸烟状况、上消化道恶性肿瘤家族史和病理分期状态,E-cadherin-160基因型及等位基因型在胃癌患者中的分布未呈现出显著性差异(P>0.05)。结论 E-cadherin C-160A多态性基因型和等位基因型与胃癌的易感性相关,E-cadherinC-160A多态性与胃癌患者吸烟状况、上消化道肿瘤家族史和术后病理分期之间无相关性。 Objective To explore the association between the E-cadherin gene promoter C-160A site with the susceptibility to gastric cancer. Methods Totally 150 patients with gastric cancer and 150 healthy controls were selected. E-cadherin -160 genotypes were examined by PCR-RFLP method. Results The overall genotype and allclotype distributions of E-cadherin gene promoter-160 site in the gastric cancer patients were significantly different from those in the healthy controls ( P 〈 0.05). Stratification analysis indicated that the frequency of E-cadherin gene promoter-160 genotypes and allelotypes in gastric cancer and control groups presented significant differences no matter the objectives were smoking or non-smoking (P〈0.05). After gastric cancer cases were stratified according to upper gastrointestinal cancer (UGIC) history or pathological staging, no significant differences were detected in the frequencies of E-cadherin genotypes between the gastric cancer patients and the healthy controls (P〉0.05). Oonclusion The E-cadherin gene promoter C-160A single nucleotide polymorphism is associated with the susceptibility to gastric cancer, but not associated with smoking status, UGIC familial history or pathological stage.
