摘要
目的探讨心力衰竭(chronic heart failure,CHF)大鼠血浆心房钠尿肽(atrial natriuretic polypeptide,ANP)水平与心肌羟脯氨酸(hydroxyproline,HYP)含量的关系。方法以腹腔注射盐酸阿霉素的方法建立大鼠心力衰竭模型(CHF组,n=10),另设正常对照(NS组,n=10)。用放射免疫法测定血浆ANP含量;经颈动脉测量左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室压力最大上升及下降速度(±LVdp/dtmax)心功能指标;病理学切片检测,采用HE染色、显微镜观察;采用氯胺T法测定心肌HYP含量。结果 CHF组大鼠较对照组,心功能指标LVSP及±LVdp/dtmax均明显下降(P<0.01),LVEDP明显升高(P<0.01),血浆ANP含量及心肌HYP含量均显著升高(P<0.01);CHF组大鼠血浆ANP含量与心肌HYP含量呈高度正相关(r=0.818,P<0.01);CHF组大鼠血浆ANP含量与心功能+LVdp/dtmax呈高度负相关(r=-0.775,P<0.05)。结论心力衰竭大鼠血浆ANP含量及心肌HYP含量均随心功能减低而显著升高,心肌HYP含量随血浆ANP水平升高而升高。心力衰竭大鼠血浆ANP可以通过反映心肌组织纤维化指标HYP的水平从而反映心肌纤维化的程度。
Objective To investigate the relationship between atrial natriuretic polypeptide(ANP) levels and myocardial hydroxyproline(HYP) contents in rats with chronic heart failure(CHF).Methods The CHF was induced by intraperitoneal injection of doxorubicin hydrochloride(ADR).Plasma ANP levels were determined by radioimmunoassay in 10 CHF rats(CHF group) and 10 control rats(NS group).Left ventricular systolic blood pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),and maximal rate of left-ventricular pressure rise and decline(±LVdp/dtmax) were measured to evaluate the cardiac function.Histopathological changes were detected by HE staining under the microscope.Myocardial HYP contents were determined by chloramine-T method.Results Compared with control group,CHF induced significant decrease in LVSP and ±LVdp/dtmax but obvious increase in LVEDP,plasma ANP levels and myocardial HYP contents(P0.01).Plasma ANP levels were positively correlated with myocardial HYP contents(r=0.818,P0.01) but negatively correlated with +LVdp/dtmax(r=-0.775,P0.01) in CHF rats.Conclusion Compared with normal rats,plasma ANP levels and myocardial HYP contents significantly increase with decreased heart function in CHF rats.Furthermore,there is a positive correlation between plasma ANP levels and myocardial HYP contents.Therefore,plasma ANP levels can reflect the degree of myocardial fibrosis in CHF rats.
