摘要
目的研究低氧条件对体外培养人胃黏膜上皮细胞株GES-1凋亡的影响。方法建立GES-1细胞体外低氧培养模型。在低氧处理后不同时间点(24 h、48 h、72 h)MTT法检测存活率、电镜观察形态变化、TUNEL染色观察细胞凋亡。结果低氧条件作用24 h、48 h、72 h后,GES-1细胞存活率均下降(P<0.05),细胞凋亡指数均升高(P<0.05),透射电镜均观察到部分凋亡细胞。结论低氧条件可明显抑制GES-1细胞生长并诱导其凋亡。
Objective To investigate the effect of hypoxia on apoptosis in human gastric epithelial cell strain-1 ( GES- 1 ). Methods Hypoxia culture model in vitro was established to test the apoptosis in GES-1 cell line. After GES-1 was cultured in hypoxia circumstance for a given period of time (24 h, 48 h, 72 h). The survival rate of cell and apoptosis were measured by MTI', electromicroscope, TUNEL staining. Results The survival rate of GES-1 was decreased (P 〈 0.05 ), but the apoptosis index was increased (P 〈 0.05 ) in hypoxia at 24 h, 48 h, 72 h. The transmission electron microscope showed some apoptosis-like morphologic changes. Conclusion Hypoxia has an obvious inhibitory effect on the cell proliferation, and increases apoptosis of GES-1.
