人参皂苷Rb1对大鼠实验性心肌缺血再灌注损伤的保护作用 被引量:8

摘要 目的观察人参皂苷Rb1对大鼠心肌缺血再灌注损伤的保护作用,并初步探讨其机制。方法在体大鼠结扎冠状动脉前降支30min,再灌注24h制备实验性心肌缺血再灌注损伤模型,人参皂苷Rb1按50和100mg.kg-1.d-1给大鼠连续灌胃7d,空白组及再灌模型组灌胃生理盐水。计算心肌梗死面积,测定心肌组织超氧化物歧化酶(SOD)活性,NO及丙二醛(MDA)含量,RT-PCR检测心肌局部eNOS表达水平。结果与再灌模型组比较,人参皂苷Rb1 50、100mg.kg-1组大鼠的MIS显著缩小(P<0.05或P<0.01),心肌MDA含量降低(P<0.05或P<0.01),SOD及NO活性增强(P<0.05或P<0.01),eNOS表达水平增加明显(P<0.05或P<0.01)。结论人参皂苷Rb1对大鼠实验性心肌缺血再灌注损伤具有明显保护作用,可能与其增强抗氧化酶活性、减少自由基对心肌的氧化损伤、增强eNOS表达等机制有关。 Objective To study the protective effect of ginsenoside Rb1 on myocardial ischemia-reperfusion in rats and its mechanisms.Methods The model of myocardial ischemia-reperfusion injury was established by ligating the anterior branch of left coronary artery for 30 min,followed by 24h reperfusion.Ginsenoside Rb1 was administrated intragastrically at the dose of 50 or 100mg·kg-1·d-1,for 7 days after the ischemia-reperfusion injury.Sham and control group were given physiological saline.The volume of myocardial infarction,the superoxide dismutase(SOD),the nitric oxide(NO),malonaldehyde(MDA) and the transcription level of endothelial nitric oxide(eNOS) in ischemia-reperfusion injured myocardium were measured.Results Compared with control group,ginsenoside Rb1 could significantly decrease the volume of myocardial infarction and the content of MDA,elevate the levels of SOD,NO and eNOS.Conclusion Ginsenoside Rb1 can protect myocardium from ischemia-reperfusion injury in rats,and the mechanisms may be related with reduction of oxygen radical genenation and elevation of of eNOS level.
