黄芪预防小鼠急性酒精性肝损伤的作用机制研究 被引量:7

摘要 目的探讨黄芪对急性酒精性肝损伤小鼠氧化应激的影响及其作用机制。方法用不同剂量黄芪(0.5g/kg和1.0g/kg)灌胃,1次/d,连续7d,然后用50%白酒灌胃(0.2ml/10g),1次/d,共3次,制备小鼠急性酒精性肝损伤模型,并取15只健康小鼠作正常对照。测定各组小鼠血浆丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)的活性;并测定肝组织中的超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)的活性及谷胱甘肽(GSH)和丙二醛(MDA)的含量。结果①模型组小鼠ALT、AST和MDA显著高于正常组(P<0.01),而SOD、GSH-PX和GSH显著低于正常组(P<0.01);②各剂量黄芪口服液能显著降低ALT和AST的活性和MDA含量(P<0.05或P<0.01);提高SOD和GSH-PX的活性和GSH含量(P<0.05或P<0.01);③黄芪两剂量组间比较:高剂量组ALT和AST的活性显著降低(P<0.05或P<0.01);而GSH-PX和GSH显著升高(P<0.05)。结论黄芪能明显减轻酒精性肝损伤,机制可能与其抗氧化应激作用有关。 Objective To investigate the effects of astragalus membranaceus on oxidative stress in mice with alcoholic liver injury and explore the protective mechanism of astragalus membranaceus for acute alcoholic liver injury.Methods The different doses of astragalus membranaceus(0.5,1.0g/kg)were intragastrically administrated,once a day for 7days.The mouse model of acute liver injury was induced by means of intragastric infusion of 50% alcohol,once a day for 3 days.The activities of alanine amino-transferase(ALT),aspartate amino-transferase(AST) in plasma,superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) in liver tissues and the contents of malondialdehyde(MDA) and glutathione(GSH) were determined.Results Compared with normal control group,the levels of ALT,AST and MDA in model control group were significantly increased(P0.01),and the levels of SOD,GSH-PX and GSH were significantly decreased(P0.01).Compared with model control group,astragalus membranaceus could obviously decrease the levels of ALT,AST and MDA and significantly increase the levels of SOD,GSH-PX and GSH(P0.05 or P0.01).Compared between the two-dose groups of astragalus membranaceus,the effects of high-dose group were more obviously and there had significant difference(P0.05 or P0.01).Conclusion Astragalus membranaceus can alleviate acute alcoholic liver injury,and the mechanism is probably related to the anti-oxidative stress.
