摘要
目的观察注射用水蛭低肽冻干粉针剂的体内抗凝、抗粘及溶栓作用。方法通过毛细管法测定各组小鼠凝血时间的差别;通过注射盐酸肾上腺素及冰水刺激造成大鼠血瘀模型,再比较空白组、模型组、对照组及高中低剂量组的血栓湿长、湿重、干重,及血流变、血沉、红细胞压积。结果注射用水蛭低肽冻干粉针剂20、40、80μg.kg-1能显著延长小鼠的凝血时间,并能显著降低血瘀大鼠的血栓湿重、湿长及干重,血沉及红细胞压积。40、80μg.kg-1能降低血瘀大鼠的全血粘度,而且剂量均较重组水蛭素低。结论自制的注射用水蛭低肽冻干粉针剂与重组水蛭素药理活性相似,具有明显的抗凝、降粘、降脂作用。
Objective To observe the esoteric anticoagulation, anti - viscosity and thrombolysis activity of Sanguisuge Low - peptide Freeze Dried Powder for Injection. Methods The deference of clotting time was determined by capillary tube method ;The blood stasis model of rat was induced by injecting adrenaline hydrochloride and giving the ice water stimula- tion, and then the thrombus length, wet and dry weight, blood rheology, blood sedimentation, hematocrit were compared a- mong the blank group,model group,control group,and the high middle low dose groups. Results 20,40 and 80 μg·kg-1 of Sanguisuge Low - peptide Freeze Dried Powder for Injection can significantly lengthen the clotting times significant of mice and decrease the wet weight of thrombus, wet length, dry weight, blood sedimentation and hematocrit of blood stasis rats. 40,80 Ixg ~ kg-1 of Sanguisuge Low- peptide Freeze Dried Powder for Injection can significantly decreased the whole blood viscosity of blood stasis rats and the concentration was lower than the recombinant hirudin. Conclusion Sanguisuge low - peptide Freeze Dried Powder for Injection had the similar effects with recombinant hirudin, and it had obvious antico- agulation, reduce viscosity and blood- fat effects.
