Expression of transforming growth factor beta 1 in gastric carcinoma tissue with Helicobacter pylori infection

摘要 目的检测幽门螺杆菌(HP)阳性和阴性胃癌患者的病变组织中转化生长因子β1(TGF-β1)的表达分布,并分析其与胃癌患者临床病理学特征的相关性。方法选取39例胃癌患者的胃镜活检组织标本,采用快速尿素酶试验(RUT)结合Giemsa染色判断HP感染。应用冰冻切片免疫组织化学染色法检测胃癌组织中TGF-β1的表达分布。结果 HP阳性者TGF-β1表达(78.6%)显著高于HP阴性者(36.4%)(P<0.05),胃癌患者HP感染与TGF-β1的表达相关。临床Ⅰ期与Ⅱ、Ⅲ、Ⅳ期患者以及浸润深度为T1与T2、T3、T4胃癌患者的癌组织中TGF-β1阳性表达差异具有统计学意义(P<0.05)。胃印戒细胞胞浆频繁出现TGF-β1强阳性染色。结论胃癌患者中HP感染与TGF-β1的表达存在相关性,TGF-β1在胃癌发生、发展中发挥着重要作用,TGF-β1可成为早期胃癌诊断、治疗和预后的分子标志,HP可能通过TGF-β1信号转导通路而在胃癌的发生、发展中起到重要作用。 Objective To detect the expression and disposition of transforming growth factor beta 1 ( TGF-β1 ) in gastric carcinoma tissue with Helicobacter pylori (HP) infection and without Helicobacter pylori infection and to analyze the correlation between the expression of TGF-β1 and the clinical pathological characteristics. Methods Total of 39 cases with gastric carcinoma were collected from gastroseopy with or without Helicobacter pylori infection were determined by rapid urease test (RUT) combined with Giemsa staining. The levels of TGF-β1 were compared by frozen section method for immunohistochemieal staining. Results The positive rate of TGF-β1 expression in HP-positive gastric carcinoma patients was significantly higher than that in HP-negative patients (P 〈 0.05 ). The TGF-β1 expression levels were significantly different among the stage Ⅰgroup and the stage Ⅱ ,Ⅲ, Ⅳ group; which was the same to T1 depth of invasion and T2, T3, T4 depth of invasion (P 〈 0.05 ). Frequently, the TGF-β1 staining was strong in gastric sinet-ring cells. Conclusions There is a significant correlation between the expression of TGF-β1 and Helicobacter pylori infection in gastric carcinoma tissue. TGF-β1 plays an important role in the pathogenesis of gastric carcinoma. It could be a molecular marker in early diagnosis, treatment and prognosis in the gastric carcinoma. Helicobacter pylori may play an important role in the development of gastric carcinoma through TGF-β1 signal pathway.
