摘要
目的:研究知母多糖对O2-.和.OH两种自由基的清除作用以及抑制蛋白酪氨酸磷酸酶PTP-1B活性的能力,为知母多糖药物及功能性食品开发提供理论依据。方法:采用Fenton体系和邻苯三酚法测定知母多糖提取物对O2-.和.OH的清除作用。对知母多糖提取物分别进行膜分离以及层析柱分离,用酶标仪测定分离所得提取物以及不同浓度下知母多糖溶液对PTP-1B活性的抑制能力。结果:知母多糖提取物具有良好体外抗氧化性以及较好的抑制PTP-1B活性的能力。结论:知母多糖提取物是一种良好的氧自由基清除剂以及PTP-1B抑制剂。
Objective : The scavenging effect and inhibitory activity of anemaran extractions on the superoxide anion free radical L ( O^-2· ), hydroxyl free radical ( ·OH) and protein tyrosine phosphatasc 1 B ( FIT - 1 B) Were investigated for the applications of anemaran extractions in drug and health foods. Methods : The Fenton reaction method and auto-oxidation of pyrogallol method were used for the scavenging of O^-2· and ·OH. After membrane and column chromatography separation, the inhibitory activity of anemaran extractions with different concentrations to PTP - 1 B was measured by enzyme-labelled microplate reader. Results : The anemaran extractions showed good antioxidant activity and inhibitory activity on protein tyrosine phosphatase 1 B in vitro. Conclu- sion:Anemaran extraction is an effective scavenger of oxygen free radicals and protein tyrosine phosphatase 1 B inhibitor.
