摘要
目的:观察外寒对小鼠肺组织BAFF和NF-κB表达的影响。方法:50只昆明小鼠随机分为5组:常温对照组、4℃7天组、4℃14天组、0℃7天组、0℃14天组,置于人工气候箱中处理后取小鼠肺脏进行病理切片,ELISA法检测IgG-S和IgG-R,实时荧光定量PCR检测BAFF的表达,Western blot法检测小鼠肺组织中NF-κB蛋白的表达情况。结果 4℃7天组、0℃7天组小鼠肺部出现明显的病理的改变,肺泡内出现大量的渗出液,各实验处理组IgG-S和IgG-R较常温对照组明显升高,差异具有显著性意义,实时荧光定量PCR检测发现各处理组BAFF的表达与常温对照组比较,明显上调,Western blot结果显示各处理组NF-κB蛋白的表达较常温对照组明显增加。结论外寒通过上调BAFF和NF-κB的表达,促发机会的获得性免疫。
Objective:To observe the effects of external cold on the BAFF and NF - KB expressions in lung tissue of mice. Methods:50 Kunming mice were divided into five groups:normal group,4℃/7d group,4℃/14d group,0℃/7d group and 0℃/ 14d group. Mice were put into different temperature - controlled boxes, then samples were collected to observe their pathological changes in pulmonary tissue, IgG - S and IgG - R were measured with ELISA. The expression of BAFF was detected by real time PCR, and NF - KB was detected by western blot. Results:Compared with the normal group, the 4℃/7d group and 0℃/7d group pulmonary tissues had obvious changes, alveoli edema could be observed. IgG - S and IgG - R of the 4℃/7d group ,4℃/14d group,0℃/7d group and 0℃/14d group were higher( P 〈 0. 05 ). The BAFF and NF- KB expressions of 4℃/7d group and 0℃/7d group were higher than that of the normal group. Conclusion:The external cold impaired lung,the defense function of lung was damaged and caused the pulmonary infection,which induced the acquired immunity and made the expressions of BAFF and NF - KB increase.
