人促红素对溃疡性结肠炎小鼠血清Th17细胞因子表达的影响 被引量:1

摘要 目的:探讨人促红素(hEPO)对溃疡性结肠炎(UC)小鼠血清Th17细胞因子的表达影响。方法:将30只BALB/C小鼠随机分为3组:正常组、DSS模型组和hEPO组,每组10只。葡聚糖硫酸钠(DSS)诱导小鼠UC模型,观测小鼠疾病活动指数评分(DAI评分),第8 d处死小鼠取病变结肠病理切片和评分,采用Elisa法检测小鼠血清IL-6和IL-17A的含量。结果:各组小鼠DAI评分、血清IL-6和IL-17A含量、结肠病理评分均较正常组高;经hEPO治疗后小鼠的各指标均下降,与DSS组比较,差异有统计学意义(P<0.05)。结论:人促红素能有效减轻UC小鼠临床症状和肠道损伤,推测hEPO抑制了血清IL-6的表达和释放,从而减少Th17细胞分化生成IL-17A和达到治疗UC的目的。 OBJECTIVE:To approach the influence of human erythropoietin on the expression of Th17 cell factor in serum of mice with ulcerative colitis.METHODS:Thirty BALB/C mice were randomly divided into normal group,DSS model group and hEPO group,10 mice in each group.UC murine was induced by dextran sodium sulfate.All mice disease activity index score(DAI score) were calculated.Mice were killed at eighth day,the colon tissues were conducted pathological section and score,serum content of IL-6 and IL-17A were assayed by Elisa kits.RESULTS:The score of DAI and histological damage score,serum IL-6 and IL-17A content were higher in model group than that in normal group(P0.05),which were significantly improved by erythropoietin treatment,the difference had statistical significant(P0.05).CONCLUSION:Human erythropoietin can effectively lessen the clinical symptom and injury of intestinal tract of mouse with UC,we supposed that hEPO inhibits the expression and release of serum IL-6,and reduces Th17 cell differentiation into IL-17A,and finally reaches to the purpose of treatment of UC.
