摘要
目的:探索加味四君子汤对肌萎缩侧索硬化症(ALS)AR2小鼠运动功能的影响。方法:采用转基因方法制作保留部分运动功能的ADAR2flox/flox小鼠为对照组(Ⅰ组),将ALS AR2小鼠随机分为空白组(Ⅱ组)、中药低剂量组(Ⅲ组)及高剂量组(Ⅳ组)。各组小鼠正常进食水,中药组分别给予灌胃低、高剂量的加味四君子汤,每日1次。每4周观察小鼠活动状况以及抓握力、爬滚筒能力等运动功能,并在第24周检测小鼠血常规、心肝肾功能。结果:各组小鼠第8周开始均有不同程度的运动功能减退,到24周为止Ⅰ组运动功能减退速度缓慢,Ⅱ组AR2小鼠从12周开始无论活动度、抓握力还是爬滚筒能力均迅速下降,而AR2小鼠中药治疗组运动功能降低程度被延缓,且与中药剂量有关;24周时Ⅱ组AR小鼠的体质量、抓握力和爬滚筒能力与其余3组比较明显降低,差异有统计学意义。各组小鼠24周时的生化检测均无异常。结论:加味四君子汤能够延缓ALS小鼠运动功能的衰退。
Objective: To investigate the effects of “Jiawei Sijunzi Decoction” on movement dysfunction of AR2 mice with amyotrophic lateral sclerosis (ALS). Methods: The ADAR2 flox/flox mouse model which remained part of motor functions was established by transgenic methods, and was taken as the control group ( group I ). The ALS AR2 mice were randomly divided into blank group ( group Ⅱ ) , low dose of Chinese medicine group ( group Ⅲ ) and high dose of Chinese medicine group (group IV ). All the mice were given normal food and water, and the mice in group m and group IV were fed intragastrically by low dose and high dose “Jiawei Sijunzi Decoction”respectively once a day. The active situation, strength of grip and other movement functions were observed every 4 weeks, and the routine blood and heart, liver and kidney functions were detected at the 24th week. Results: All the mice began to appear different degrees of motion hypofunction from the 8th week, the progress of motion hypofunction was the lowest in group I till the 24th week, the motion functions of mice in group Ⅱ decreased quickly, and the motion functions of mice in group m and group IV were relieved related to the dose of the Chinese medicine. Compared with the other groups, the body weight and motion functions of AR mice in group Ⅱ significantly decreased with statistical difference at the 24th week. There were no differences on biochemical indexes among all the groups. Conclusion: “Jiawei Sijunzi Decoction”can delay the motion hypofunction of ALS mice.
