生地免煎颗粒对大鼠脑缺血后Nogo-A mRNA表达的干预作用

Intervention of “Shengdi Extract Granule” on Expression of Nogo-A mRNA in Rats Following Cerebral Ischemia

目的:观察生地免煎颗粒对神经生长抑制因子勿动蛋白(Nogo-A)mRNA表达的影响。方法:将雄性健康SD大鼠分为用药组(A组)、模型组(B组)和假手术组(C组),采用生地免煎颗粒溶液灌胃的方式作用于大脑中动脉闭塞(MCAO)模型大鼠,分别在造模后第3、7、14天3个时间点采用实时荧光定量PCR方法观察Nogo-A mRNA的表达。结果:经MCAO造模后,模型组Nogo-A mRNA的表达较假手术组明显下调,在第3天和第14天存在显著性差异(P<0.01);经生地免煎颗粒溶液灌胃后,用药组Nogo-A mRNA的表达在3个时间点均低于假手术组(P<0.05),在第7天时间点上明显低于模型组(P<0.05),其余时间点未见显著性差异。结论:MCAO造模后,大鼠脑内Nogo-A mRNA的表达有所下调,有利于缺血性脑损伤后的神经再生;生地免煎颗粒能够在特定时间段进一步抑制Nogo-A mRNA的表达,从而更有利于缺血性脑损伤后的神经再生。

Objective： To observe the effects of “Shengdi Extract Granule” on the expression of Nogo-A mRNA in rats following cerebral ischemia. Methods： The SD male rats were randomly divided into treatment group （group A）, model group （group B） and sham-operated group （group C）. The MCAO model rats were administered intragastfically with “Shengdi Extract Granule”. The expression of Nogo-A mRNA were detected by real-time qPCR on the third, seventh, fourteenth days after modeling. Results ： The expression of Nogo-A mRNA in model group was lower than that in sham-operated group, with statistical differences at the third and fourteenth days between two groups （P 〈0.01 ） ; The expression of Nogo-A mRNA in treatment group was lower than that in sham-operated group at the three times （P 〈0.05 ）, and was lower than that in model group at the seventh day （P 〈0.05 ）. Conclusion： The expression of Nogo-A mRNA in MCAO model rats decreases, and this is beneficial to the nerve generation after cerebral ischemic. “Shengdi Extract Granule” can further inhibit the expression of Nogo-A mRNA and contribute to the nerve generation.
