卡那霉素和草铵膦对不同基因型大豆胚尖不定芽诱导的影响 被引量:4

Effect of Knamycin and Glufosinate on Adventitious Buds Induction from Embryonic Tip of Different Genotype Soybean

摘要 以合丰35、黑农44和吉林35的胚尖为外植体,分别考察了不同浓度的卡那霉素和草铵膦对不同基因型大豆胚尖不定芽诱导的影响。结果表明,不定芽诱导率合丰35和吉林35在卡那霉素100 mg.L-1时与对照差异不显著,黑农44在100 mg.L-1处理时显著低于对照;芽数吉林35在卡那霉素100 mg.L-1时显著低于对照并高于其他浓度,而合丰35与黑农44在100 mg.L-1时芽数与对照差异不显著;3种基因型大豆胚尖不定芽伸长均在卡那霉素浓度为100 mg.L-1时受到显著抑制。草铵膦浓度在0.2~1.2 mg.L-1时对合丰35和吉林35的芽数没有影响,但黑农44在1.0 mg.L-1时芽数开始显著低于对照;芽长合丰35、黑农44和吉林35分别在0.6、0.2和0.2 mg.L-1时显著低于对照。因此,合丰35、黑农44和吉林35的适宜卡那霉素的筛选浓度为100 mg.L-1,草铵膦浓度分别为0.6、0.2和0.2mg.L-1。 Embryonic tips of ttefeng35 ,Heinong44 and Jilin35 were used as the explants to investigate the effects of kanamy- cin and glufosinate on the adventitious bud induction. Under 100 mg·L-1 kanamycin, the adventitious bud induction rate of Hefeng35 and Jilin35 were not significantly different compared with control, while Heinong44 was significantly lower than con- trol ; bud number of Jilin35 was lower than control and higher than other concentrations, Hefeng35 and Heinong44 were not dif- ferent compared with control;the shoot length of 3 genotypes were significantly inhibited by 100 mg· L-1 kanamycin. Glufosi- nate at the concentration from 0.2 to 1.2 mg· L- 1 had no intluence on the number of adventitious buds for Hefeng35 and Ji- lin35, but Heinong44 reduced conspicuously at the concentration of 1.0 mg· L - 1. Both Heinong 44 and Jilin 35 were inhibited by 0.2 mg·L - 1 glufosinate at length of buds while Hefeng35 was inhibited by 0.6 mg·L - 1 glufosinate. Results suggest the suit- able screening concentration of kanamycin for Hefeng35, Heinong44 and Jilin35 were 100 mg· L -1, and glufosinate for three genotypes were 0.6,0.2 and 0.2 mg·L-1 ,respectively.
