新型HA联合BMSCs修复大鼠胫骨缺损的实验研究

摘要 目的 BMSCs联合新型HA修复大鼠胫骨缺损,为骨组织工程提供新的数据。方法首先分离培养大鼠BMSCs。制备新型HA支架,将细胞与支架进行复合移植给大鼠5mm胫骨离断性缺损区,于2、8和12w时分别处死动物。X线检测缺损区骨修复情况,组织学观察成骨情况。结果制备的HA支架孔隙丰富,均匀。术后12w,支架大部分降解,无炎性反应,可见大量板层骨样结构。X线显示缺损区可见新生骨痂且新骨密度增高,形成桥接。结论新型HA与BMSCs复合修复大鼠5mm胫骨缺损效果良好。 Objective To repair tibia bulk defect with BMSCs and hydroxyapatit composit in rats, and provide new data for bone tissue engineering. Methods First the experimrnt isolated and cultured rat BMSCs, and prepared hydroxyapatite, then the BMSCs and scaffold composite was transplanted into 5 mm tibia defect area in rats, 2 w, 8 w and 12 w later the rats were sacrificed. X-ray detected the defect bone repair, and and osteogenesis was observed by histologic method. Results The hydroxyapatite scaffold had abundant pores. After 12 w of transplantation, the scaffold degraded mostly without inflammation, and mass mature lamellar bone was formed. X-ray showed the defect area had new bone callus and new bone density increased. Conclusions BMSCs and hydroxyapatite composite will have better effect in repairing 5 mm tibia bulk defect in rats.
