摘要 芬戈莫德(fingolimod,FTY720)是冬虫夏草中提取的有效成分多球壳菌素(myriocin,ISP-1)通过结构改造而合成的一种新型免疫抑制剂。与传统的免疫抑制剂不同,它不影响淋巴细胞的活化和增殖,主要作用于细胞表面的鞘氨醇-1-磷酸受体(S1PR)来发挥免疫抑制和免疫调控作用。本文介绍FTY720对T、B淋巴细胞和天然免疫细胞(树突状细胞、巨噬细胞、自然杀伤细胞、自然杀伤性T细胞)的分布和细胞因子产生的影响,探讨FTY720对炎症的免疫抑制和免疫调控作用;对FTY720在自身免疫病、移植物抗宿主病和肿瘤治疗等方面的研究进展,FTY720的副作用及可能的解决方案做了简要总结。 Fingolimod(FTY720),a novel immunosuppressive agent,is synthesized by modification of myriocin(ISP-1) from a traditional Chinese herbal medicine Isaclaria sinclrii.Unlike the traditional immunosuppressive agents,fingolimod exerts immunosuppressive and immunoregulatory functions mainly through interaction with shhingosine-1-phosphate receptors(S1PR) on the cell surface without affecting activation and proliferation of lymphocytes.This article briefly reviews the effects of fingolimod on the distribution and cytokine production of lymphocytes as well as natural immune cells(dendritic cell,macrophage,natural killer cell and natural killer T cell),suggesting the immunosuppression and immune regulation functions of fingolimod in inflammation.Then the latest research progress about the therapeutic application of fingolimod in autoimmune diseases,graft-versus-host diseases and tumors,the adverse effects of fingolimod and possible solutions are summarized.
