摘要 以红叶石楠扦插苗的茎尖作为外植体,研究了不同消毒剂的消毒灭菌效果,以及不同培养基、不同激素组合对茎尖生长扩繁的影响。试验结果表明:(1)红叶石楠茎尖外植体最佳消毒方法为用0.1%的升汞消毒处理4 min,存活率达56.7%;(2)最合适的基础培养基为MS培养基;(3)最佳的增殖培养基为MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,愈伤诱导率为52.0%,增殖系数为4.1;(4)最佳的生根培养基为1/2MS+IBA 0.5 mg/L,生根率达55%,平均根数为2.2。 Taking the stem-apex segments of Photinia fraseri as explants,the sterilization effect of different sterilants and the influence of different culture media and hormone combinations on growth and propagation of stem-apex were studied.The results showed as follows:(1) The best sterilization method for Photinia fraseri explants was treating with 0.1% HgCl2 for 4 min,and the survival rate reached 56.7%.(2) The optimal culture medium was MS medium.(3) The optimal proliferation medium was MS+6-BA 2.0 mg/L+NAA 0.2 mg/L;the callus induction rate was 52.0% and the multiplication coefficient was 3.3.(4) The most suitable rooting medium was 1/2MS+IBA 0.5 mg/L;the rooting rate reached 55.0%,and the average number of roots was 2.2.
