摘要
目的:探讨丝胶对2型糖尿病大鼠海马神经型一氧化氮合酶(nNOS)表达的作用。方法:小剂量链脲佐菌素连续腹腔注射建立2型糖尿病大鼠模型,并给予丝胶灌胃。SP免疫组化染色观察海马神经元nNOS的表达。结果:nNOS免疫阳性产物呈棕黄色细腻颗粒状,位于海马神经元的胞浆。与正常对照组大鼠相比,糖尿病模型组大鼠海马神经元nNOS的表达明显升高(P<0.01),丝胶治疗组大鼠海马神经元nNOS的表达明显低于糖尿病模型组(P<0.01)。结论:丝胶可通过下调糖尿病大鼠海马nNOS的表达,减轻过量一氧化氮导致的神经毒性,发挥对海马的保护作用。
Objective:To investigate the effects of sericine on neuronal niic oxide synthase (nNOS) expression in hippocampus of type 2 diabetes mellitus rats.Methods:The type 2 diabetes mellitus rats model was established by continuous peritoneal injection of small dose streptozotocin, and then the model rats were lavaged with sericine. SP immunohistochemical stain was used to observe nNOS expression in neurons of hippocampus.Resulls:The nNOS positive products were exquisite granules that located in cytoplasm of neurons of hippocampus. Compared with rats in normal control group, the nNOS expression in neurons ofhipppocampus of rats in model group were obviously increased (P〈 0.01 ). While, the nNOS expression in neurons of hi--us of rats in sericine Ireatment group were obviously lower than that of rats in model group (P〈 0.01). Conclusions: Sericine may play a role in protecting hippocampus during diabetes mellitus, the mechanism maybe lighten the nemrotoxicity induced by overdose nitric oxide by down regulating of nNOS expression in hippocampus.
