东莨菪碱致学习记忆损伤大鼠不同脑区PREG含量的变化

摘要 目的:观察东莨菪碱致记忆损伤大鼠不同脑区孕烯醇酮(PREG)含量的变化。方法:东莨菪碱腹腔注射制备大鼠学习记忆损伤模型,Morris水迷宫检测大鼠的习记忆能力,高效液相色谱-质谱法检测大鼠额叶皮质及海马PREG的含量。结果:模型大鼠的学习记忆能力显著下降;与对照组大鼠比较,模型组大鼠额叶皮质PREG的含量显著下降(P<0.05),海马PREG的含量无明显变化。结论:东莨菪碱致记忆损伤模型大鼠额叶皮质PREG的含量显著降低,提示东莨菪碱可能通过影响PREG的合成损伤大鼠的学习记忆能力。 Objective:To observe the changes of pregnenolone (PREG) content in different brain region of sco ent rats model was made by peritoneal injection of scopolamine. The learning and memory ability of rats were detected with Morris water maze; the PREG content in cortex of frontal lobe and hippocampus were detected by high performance liquid chromatography-mass spectrometry (HPLC-MS).ResulIs:The learning and memory ability of model rats decreased significantly compared with control rats. Compared with conlrol rats, file PREG contents in cortex of tiontal lobe decreased significantly (P〈 0.05);While PREG content in rats' hi--pus had no significantly changed.Conclusions: The PREG content in cortex of frontal lobe of scopolamine-induced learning and memory impainnent rats decreased significantly, which infers that scopolamine may impair the rat's learning and memory ability by influenceing the synthesis of PREG.
