摘要
探讨天然促智药复方——补肾益智方对基底前脑神经元型一氧化氮合酶(nitric oxidesynthase1, NOS1)表达的影响。方法:用鹅膏蕈酸(Ibotenic acid,IBO)损毁老年大鼠双侧基底核作为老年性痴呆(Alzheimer’s disease, AD)模型,治疗组行补肾益智方灌胃,用药对照组用石杉碱甲(Hup A)灌胃,正常对照组为青年组和老年正常组。各组Morris水迷宫检测,之后取各组受试大鼠脑组织冠状切片进行NOS1免疫细胞化学染色。结果:逃避潜伏期中药组大鼠较模型组、Hup、 A组明显缩短。 NOS1阳性细胞数中药组较模型组、Hup A组明显增加,明显低于青年组和老年正常组。灰度值中药组较青年组、老年正常组明显降低,斜角带水平支( horizonal diagonal band of Broca, hDB)较模型组有明显增加。胞体面积中药组较模型组、Hup A组显著增加。结论:补肾益智方可增加基底前脑 NOS1表达。
Objective: To explore the effect of Bushenyizhi Decoction, a Chinese compound prescription composed of natural nootropic drugs and indicated for tonifying the kidney and strengthening the brain), on the expression of nitric oxide synthase 1 (NOS1 ) in the hasal forebrain. Method: The AD model (model with Alzheimer's disease) was created from an old rat whose double sides of nuclei basales were damaged by ibotenic acid (IBO). The treating group was poured with Bushenyizhi Decoction, and the control group with Hup A. The normal controls were composed of the young and the normal aged. Every group was tested with Morris water maze, and then the frontal sections of brain tissues were stained immunocytochemically with NOS1. Result: The escape latency of the treating group was significantly shorter than that of AD model and Hup A group. NOS1 -- ir neuron number of the treating group was siginificantly greater than that of AD model and Hup A group, and markedly smaller than that of the young and the aged. Compared with the model, the gray level of NOS1 -- ir neuron in horizontal diagonal band of Broca (hDB) of the treating group increased remarkably. The somata area of the treation group was much larger than that of the model and Hup A group. Conclusion: Bushenyizhi Decocting can increase the expression of NOS1 in the basal forebrain.
