RNA干扰技术在肿瘤基因治疗中的研究现状 被引量:11

摘要 RNA干扰(RNAinterference,RNAi)技术是利用双链RNA(double-strandedRNA,dsRNA),高效、特异性降解细胞内同源信使RNA(messengerRNA,mRNA),从而阻断特定基因表达,使细胞出现靶基因缺失的表型。1998年Fire等[1]首次应用RNAi技术将dsRNA正义链与反义链的混合物注入线虫体内,结果发现注入dsRNA比单独注入单链RNA引起的基因沉默要强得多,并可导致子一代同源基因的沉默。
