摘要 目的观察雷公藤内酯醇对鼻息肉培养组织炎症因子表达的影响。方法将鼻息肉组织体外无菌培养,加入组胺和花生四烯酸作为刺激物,观察雷公藤内酯醇在不同浓度(12.5~100 nmol/L)和不同作用时间(0.5 h,1 h,2 h,4 h,8 h,16 h)的状态下对鼻息肉释放炎症因子IL-3、IL-5、IL-8和TNF-α的抑制效应;同时,以相同浓度的地塞米松为对照,观察雷公藤内酯醇对炎症因子的抑制效应。结果雷公藤内酯醇可显著抑制组胺、花生四烯酸刺激的鼻息肉组织分泌IL-3、IL-5、IL-8和TNF-α(P<0.01),抑制效应与雷公藤内酯醇具有剂量和时间依赖关系,其中尤以对IL-8的抑制效应最明显;在相同浓度的作用条件下,雷公藤内酯醇对鼻息肉表达IL-3、IL-5和TNF-α抑制效应低于地塞米松(P<0.01),而对IL-8的抑制效应则与地塞米松相似(P>0.05)。结论雷公藤内酯醇具有抑制鼻息肉组织释放炎症因子的作用。 In this study, we aim to explore the effects of triptolide on inflammatory response factors of human nasal polyp tissue cultured in vitro. Human nasal polyp tissues were primarily cultured in vitro and then induced byhistamine and arachidonic acid. Different concentrations of triptolide (12.5-100 nmol/L) were added to the tissue, and cultured for different times (0.5 h, 1 h, 2 h, 4 h, 8 h, and 16 h). Meanwhile, the control group was treated withequivalent concentration of dexamethasone. Then ELIAS assay was performed to assess the variation of IL-3, IL-5, IL-8 and TNF-α which were secreted in the tissue. The results showed that triptolide had significantly inhibitoryeffect on the levels of IL-3, IL-5, IL-8 and TNF-α which were induced by histamine and arachidonic acid in human nasal polyp tissue cultured in vitro (P〈 0.01). Furthermore, the inhibitory effect appeared dose- and time-dependent, and demonstrated a particularly significant rate on the level of IL-8. In the same concentration of action condition, the inhibitory effect on the IL-3, IL-5, and TNF-α of triptolide was inferior to the dexamethasonetreatment group (P〈 0.01), but a similar inhibitory effect on the level of IL-8 compared with the dexamethasone treatment group (P 〉 0.05). In conclusion, we preliminary confirmed that triptolide could inhibit the inflammatoryresponse factors released by the human nasal polyp tissue.
