SREBP-1c、HSL和ATGL mRNA在不同体重指数人群皮下和大网膜脂肪组织中的表达 被引量:2

Expressions of SREBP-1c,ATGL and HSL in subcutaneous and omental adipose tissues of people with different BMIs

摘要 目的探讨固醇调节元件结合蛋白1c(SREBP-1c)、激素敏感脂肪酶(HSL)和脂肪组织甘油三酯脂肪酶(ATGL)在不同体重指数(BMI)人群的皮下脂肪组织(SAT)和大网膜脂肪组织(OAT)中的mRNA表达。方法按BMI将82例患者分为超重组(A组,35例,BMI 24-27.9)、肥胖组(B组,27例,BMI≥28)和正常体重组(C组,20例,BMI 18.5-23.9)。检测三组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL)、高密度脂蛋白胆固醇(HDL);半定量RT-PCR方法检测三组腹部SAT和OAT中SREBP-1c、HSL、ATGL 3个基因的mRNA表达水平。结果A、B组的血脂异常率高于C组(42.86%、51.86%vs.10.00%)(P<0.05)。在SAT中,B组SREBP-1cmRNA表达高于C组(P<0.05);A、B组的HSL和ATGL mRNA表达水平低于C组(P<0.05)。在OAT中,A、B组的SREBP-1c、ATGL mRNA表达高于C组,而HSL mRNA表达则低于C组(P<0.05)。结论超重和肥胖人群SAT和OAT的脂代谢基因表达水平紊乱,可能是导致其血脂异常率升高、更易罹患内分泌代谢及心血管疾病的机制之一。 Objective To investigate the mRNA expression levels of sterol regulatory element binding transcription factor-1c(SREBP-1c),hormone sensitive lipase(HSL) and adipose triglyceride lipase(ATGL) in subcutaneous adipose tissues(SAT) and omental adipose tissues(OAT) of people with different body mass indexes(BMIs).Methods According to BMI,82 patients were divided into 3 groups of A(35 cases,BMI 24-27.9),B(27 cases,BMI≥28) and C(20 cases,BMI 18.5-23.9).The mRNA expressions of SREBP-1c,ATGL and HSL were detected by RT-PCR.Serum TG,TC,LDL-C and HDL-C were measured.Results The rates of dyslipidemia in groups of A and B were higher than those in group C(42.86% and 51.86% vs.10.00%)(P0.05).In SAT,SREBP-1c mRNA expression was higher in group B than that in group C(P0.05) and HSL and ATGL mRNA expressions were lower in groups of A and B than those in group C(P0.05).In OAT,SREBP-1c and ATGL mRNA expressions were higher,but HSL mRNA expression was lower,in groups of A and B than those in group C(P0.05).Conclusion The lipid metabolism in SAT and OAT of the overweight and obesity people is in disorder,which may be one of the mechanisms for them being more susceptive to dyslipidemia,endocrine,metabolic and cardiovascular diseases.
