牛磺酸对染矽尘大鼠肺组织炎性细胞影响 被引量:6

Effects of taurine on lung inflammatory cells in rats exposed to silica

摘要 目的探讨牛磺酸对染矽尘大鼠肺组织炎性细胞趋化的影响。方法 Wistar大鼠144只,随机分为对照组、染矽尘组和牛磺酸组,后2组气管内注入质量浓度为50 g/L的矽尘混悬液,牛磺酸组在实验开始前3 d即给予含质量分数为2.5%的牛磺酸饲料持续喂养,对照组气管内注入等体积灭菌生理氯化钠溶液。偏振光显微镜观察肺纤维化发生情况,行在体全肺灌洗术,血细胞计数仪测定支气管肺泡灌洗液细胞数。结果牛磺酸组第7、14、21和28天炎性细胞总数均低于染矽尘组,差异有统计学意义(P<0.05)。牛磺酸组第14、21天巨噬细胞数低于染矽尘组,差异有统计学意义(P<0.05)。牛磺酸组第21天中性粒细胞数低于染矽尘组,差异有统计学意义(P<0.05)。牛磺酸组第14、21天淋巴细胞数均低于染矽尘组,差异有统计学意义(P<0.05)。结论牛磺酸对矽尘诱导大鼠肺组织巨噬细胞、中性粒细胞、淋巴细胞的趋化具有抑制作用。 Objective To evaluate the effects of taurine in diet on lung inflammatory cells in rats exposed to silica. Methods Animal models were established by direct tracheal instillation of silica into rat lungs exposed surgically. The formation of lung fibrosis was detected with polarization microscopy. The whole lung was lavaged in vivo. Inflammatory cell counts in bronchoalveo- lar lavage fluid (BALF) from rats were estimated by eytometer. Results Taurine treatment significantly decreased the silica-ele- vated numbers of total inflammatory cells and lymphocytes at the 1st, 3rd, 7th, 14th, 21st and 28th day, showing obviously a statistical significance at the 7th, 14th, 21st and 28th clay for total inflammatory cells (P 〈0. 05) and at the 14th and 21st day for lymphocytes (P 〈 0. 05 ) respectively. Macrophages and neutrophils in BALF were decreased at the 3rd, 7th, 14th, 21st and 28th day in taurine-treated rats compared with those of the silica-treated group, with an obvious statistical significance at the 14th and the 21 st day for macrophages and the 21 th day for neutrophils (P 〈 0.05) respectively. Conclusion Taurine treatment can effectively attenuate the inflammatory cells in the lung of rats exposed to silica in the study.
