摘要 锌指蛋白核酸酶(Zinc Finger Nuclease,ZFN)是一种人工合成酶,含有锌指蛋白和FokⅠ核酸内切酶的剪切结构域,因其能特异性识别并切割DNA序列及其可设计性而被用基因定点突变和外源基因定点整合。这项技术已应用于转基因动物研究中。本文综述了锌指核酸酶技术在转基因研究中的应用进展,并对锌指核酸酶技术的应用前景进行了展望。 Zinc finger nuclease (ZFN) is a kind of artificial synthesis containing zinc finger protein and the shear structure domain of Fok I endonuclease. Because it can specifically identify and cleave DNA se- quences and easily be designed, it is used for the site - directed genetic mutations and integration of exogenous genes. This technology has been applied to the research of transgenic animals. In this paper, the research pro- gress of zinc finger nuclease technology and its application were reviewed, and its application prospect was also forecasted.
