向日葵下胚轴遗传转化影响因素的研究 被引量:1

摘要 以NC208、HA300、诺杂212三个基因型向日葵下胚轴为材料,以带有目的基因Psy的根癌农杆菌EHA101进行转化,对影响转化的几个因素进行了研究。研究结果表明:转化受体筛选剂Hyg使用浓度为10mg/L较适宜;共培养时间3d为宜;侵染时间10min为宜;重悬液浓度A600以0.6为佳;PCR分子检测证明,目的基因Psy已整合进向日葵基因组中。 The hypocotyls of three different genotypes of sunflower (NC208, HA300 and Nuoza212) were transformed with Agrobracterium tumefaciens EHA101 carrying the target Psy gene. Meanwhile the factors affecting transformation were studied. The results were as follows, the suitable concentration of Hyg wes 10rag/L; the suitable time of co-culture was 3 days; the suitable time for infection was 10 mi- nutes; the suitable range of A600 was 0.6. PCR analysis showed that the target Psy gene could have inte- grated into the genome of sunflower.
