血府逐瘀胶囊预处理对大鼠脑缺血再灌注后病理学改变及神经细胞凋亡的影响 被引量:5

The Effects of Xuefuzhuyu Capsule Preconditioning on Histopathological Changes and Neuronal Apoptosis after Focal Cerebral Ischemia-Reperfusion in Rats

摘要 目的观察血府逐瘀胶囊预处理对SD大鼠脑缺血再灌注后组织病理学改变及神经细胞凋亡的影响,探讨其对脑缺血再灌注保护作用的可能机制。方法将138只大鼠随机分为正常组、假手术组、模型(缺血再灌注)组、复方丹参片组、血府逐瘀胶囊A组、血府逐瘀胶囊B组6组,每组23只。采用文献报道的线栓法对SD大鼠大脑中动脉梗塞模型(MCAO),于缺血2h后再灌注12h、24h、72h。观察血府逐瘀胶囊预处理对大鼠脑缺血再灌注后神经功能、脑梗死面积和病理形态学的影响。采用TUNEL凋亡法检测分析脑缺血半暗带和梗死灶中心区凋亡阳性细胞的数目。结果血府逐瘀胶囊预处理可缓解脑缺血再灌注引起的大鼠行为改变,不同程度的降低实验性脑缺血大鼠的神经功能评分(P<0.05)、并可减小脑梗死面积(P<0.05)以及减轻脑组织病理形态改变;血府逐瘀胶囊各组大鼠梗死灶面积减小,脑组织缺血半暗带凋亡细胞阳性数目减少,而梗死灶中心凋亡细胞阳性数目增多。结论血府逐瘀胶囊预处理可能通过调节脑缺血再灌注后神经细胞凋亡而促进脑损伤修复及重塑,从而发挥脑保护作用。 Objective To investigate the impact of Xuefuzhuyu capsule preconditioning on histopatho- logical changes and apoptosis of neuronal cells after focal cerebral ischemia-reperfusion and study the neu- roprotection mechanism of Xuefuzhuyu capsule. Methods 138 SD rats were randomly divided into six groups: normal (n :23) sham-operated group (n :23) ; model (ischemia and reperfusion) group (n : 23); Fufangdanshen pill group (n :23); Xuefuzhuyu capsule A group (n :23); Xuefuzhuyu capsule B group (n =23) ; Cerebral ischemia-reperfusion injury was induced in 2 h of middle cerebral artery occlu- sion (MCAO) and followed by 12 h, 24 h, 72 h reperfusion. Cerebral ischemic model was made in each group for observation of effects of Xuefuzhuyu capsule preconditioning on the neurological function, cere- bral infarction size and pathomorphology of brain tissues. The TUNEL was used to measure number of neuronal apoptosis positive cells. Results As compared with those of model group, Xuefuzhuyu capsule preconditioning decreased the score of neurological function, cerebral infarction size, and at the same time, Xuefuzhuyu capsule could lessen the degree of pathomorphological change of brain tissues (P〈0. 05). Compared with that of model group, the number of neuronal apoptotic positive cells was less a- round the necrotic area in the Xuefuzhuyu capsule group, and the number of neuronal apoptotic positive cells increased in infarction area in Xuefuzhuyu capsule group. These changes could promote the ischemic neuron to revive and become more plastic. Conclusion Xuefuzhuyu capsule preconditioning could accom- modate neuronal apoptosis after the focal cerebral ischemia-reoerfusion.
