离体大鼠心肌缺血-再灌注损伤的电子自旋共振再观察

摘要 本文通过对于大鼠离体心肌缺血-再灌注的电子自旋共振(ESR)研究,观察到Mn(Ⅱ)在大鼠心肌组织中的存在,并发现过渡金属离子与心肌细胞中产生的超氧自由基的形成与消灭有着密切关系,提出超氧自由基在组织中可能以结合态的形式存在着。并通过对在液氮下心肌组织ESR谱变化的观察,认为即使在液氮“冷冻状态”下也不能完全终止细胞内的反应,提出利用液氮保存生物自由基样品,然后进行集中检测的方法是不可取的。 The isolated rat ventricle myocardium in postischemic reperfusion injury was studied by electronspin resonance spectroscopy (ESR) at 77K. The relationship between transition metal ions and the for-mation and inhibition of superoxide free radical in reperfused rat myocardium has been found as di-scovering Mn (Ⅱ)in rat heart. The results showed that the reaction about superoxide radical in cellcould not be entirely broken off although kept it at liquid nitrogen temperature. And the method thatfree radical living samples were stored and accumulated in liquid nitrogen until measurement by ESRis unfavourable. From sampling to finishing detection at 77K should be assured of keeping less thanone hour. It is suggested that superoxide radical exists in combination states in tissues.
